Freenome provides users with innovative DNA detection methods. DNA sequencing without biopsy
In the past, DNA sequencing requires surgery to remove a living tissue from the human body for testing. It is difficult for people who are weak or sick or have poor physical condition to withstand this pain and recovery process. Unlike Freenome, their technology only requires data analysis from the patient's body fluids.
Because the genetic material of Freenome has always existed in the blood, it is the product of trillions of cells in the human body, almost everywhere. Therefore, it is only necessary to detect a small amount of blood to complete the detection of diseases such as cancer, and it is no longer necessary to cut the living slices from the human organs.
Large GWAS uses the 23andMe data to discover 15 depression loci
Gene testing company 23andMe collaborated with pharmaceutical giant Pfizer to study the DNA of more than 450,000 customers and found genetic clues to depression. The study identified 15 genomic regions associated with high depression risk. 23andMe has sold more than $1 million in genetic testing kits for $199. The product is primarily used for entertainment purposes, such as discovering the tester's ethnic background. But more than half of the clients agreed to a more in-depth analysis of their DNA and answered the health questionnaire. Through the survey, 23andMe learned that there are 141,000 customers diagnosed with depression. In the study, another 337,000 customers who did not report depression were compared as control groups.
New targets for the treatment of small cell lung cancer were discovered
Scientists from the Southwestern Medical Center have found that ASCL1 protein is essential for the development of small cell lung cancer. If the ASCL1 gene is knocked out, the mice will not develop this malignant tumor. This breakthrough advancement, published in the recent Cell publication, CellReports, shows the potential of ASCL1 as a new therapeutic target for small cell lung cancer.
After analyzing human-derived small cell lung cancer cell lines, the researchers discovered the heterogeneity of such cells: some cancer cells express ASCL1 protein, while others express NEUROD1 protein, both of which maintain humanity. The survival and cancerous behavioral characteristics of small cell lung cancer cell lines play a key role. Both ASCL1 and NEUROD1 are transcription factors that regulate the expression of many different genes involved in normal neuronal function. In addition, ASCL1 regulates the expression of many proteins in the Notch signaling pathway. However, these two proteins also stimulate transcription of protooncogenes, where ASCL1 protein activates MYCL1, RET, SOX2, and NFIB, while NEUROD1 activates MYC.
In mice, only ASCL1 but not NEUROD1 is present in pulmonary neuroendocrine tumors. When the ASCL1 gene is knocked out, the mouse model of small cell lung cancer does not develop tumors, and the knockout of the NEUROD1 gene does not.
